T2005-118 A rapid diagnostic test for Thrombotic Thrombocytopenic Purpura.

The Need

Thrombotic Thrombocytopenic Purpura (TTP) is a life-threatening microangiopathic thrombotic disorder that requires immediate diagnosis and emergency plasmapheresis therapy to prevent organ-system failure(s). Untreated TTP has a high mortality rate, but rapid diagnosis and plasma exchange therapy yields a positive prognosis. Currently, the most common way to diagnose TTP is to perform a number of tests on a patient's blood sample, and completion of these tests can take hours to days. A Western blot research test was developed, but it is cumbersome, time-consuming, and inadequate for clinical application. To diagnose TTP, a single accurate and efficient test must be developed for clinical applications.

The Technology

Researchers at The Ohio State University, led by Dr. Haifeng Wu, developed a diagnostic test for TTP using surface enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS). The test uses quantitative proteomic analysis of protein cleaved by ADAMTS-13, a metalloprotease enzyme vital to degrading von Willebrand factors (vWF) that lead to the development of TTP.
